Mechanical cell damage thresholds are important parameters for numerical models that investigate cell and tissue pathologies and disease. Our objective is to investigate chondrocyte damage thresholds as a part of the NIH project 'Predicting Cell Deformation from Body Level Mechanical Loads'. This NIH project aims to model synovial joints at multiple scales, i.e. from body to joint to tissue to cell scales, and vice versa.
We therefore aim to relate chondrocyte damage to chondrocyte deformations, and to investigate which mechanical parameter at the cell level (e.g. strain energy density, maximum shear, maximum principal strain) predicts chondrocyte damage best. A 3D culture system was used that can be subjected to a wide range of deformations on the stage of a confocal microscope with a custom made experimental set-up.
The deformed 3D culture was visualized at the microscope with a low magnification to quantify time- and space-resolved chondrocyte death. A higher magnification was used to visualise individual cells in the 3D culture, and to quantify cell shape and deformation. Finite element modeling was used to determine the average local strains of the chondrocytes in the low magnification cell damage studies.
The experiments have shown that chondrocyte sensitivity to mechanical deformation strongly depends on the culture period. Damage thresholds for young cells in culture (less than 2 days) are much lower than for cells are cultured for a longer period of time. This suggests a protective role of the extracellular matrix that is being formed by the chondrocytes during culture.
